I have a PIX 506 and the software is 5.1(2), I would like to upgrade to 6.0 or later, How much does it cost or where can I get the file?
Besides, I have DES and 3DES enabled, do I need to upgrade the activation key as well?

You'll have to purchase at least a 1 year SmartNet matintenance contract. Once you have that, you can register on Cisco website for a CCO account and then you can download the latest 6.3(4)
Since you already have the 3DES activation key, you will not need to upgrade the key again.
You can get a maint package from somplace like CDW  http://www.cdw.com

Should I need to reconfigure the PIX or the configuration will be kept during the upgrade?

The configuration will not be changed. 6.3x is fully backward compatible, but you should be aware that all "conduit" commands should be replaced with access-lists at some point. They still work, though...

*always* backup your config first, though, just in case....

The software upgrade is just that - a one-time upgrade, and if it is referencing user licensing, then it is for the PIX 501 ONLY. The 506 does not use user licensing. The 501 has 10 or 50 user license. This upgrade is probably an upgrade to the number of users for a 501.
All you need is a 1 year smartnet for the 506.
Original 506 is smartnet package category 4  $275 8x5xnbd
Later 506e is smartnet package category 2  $130 8x5xnbd
You may have found this:
PIX Software one-time upgrade for nonsupport customers (Cisco part #: PIX-SW-UPGRADE=)      $1,000.00
Not sure what that would be for....or where you would buy it..

Forgot to ask, if something goes wrong during the upgrade, is it possible to fallback to the current system version ? (I don't have the current software rom).

Yes, you can "save" your current system file just in case.
Set yourself up a TFTP server on a PC
On the PIX, copy the flash to the tftp server:
pix#copy flash tftp

Once it's on the tftp server, if you fail on the upgrade, then you have the image files..
