Transparent Images

I have a project where I want to see an image behind a filled in shape to when another shape in moved in front of it.
 Specifically, there is a rectangle that is filled on, and i want the part of it where there is a circle to be transparent.

Any other ideas? I know this a complicated question, I can use DirectX if that is necessary.
