I need to connect 4 X LCD displays to a single PC. I have done 2 X before, but need to know if it is possible to run 2 X PCI express cards on the same PC to provide 4 display with windows across them all?

The PC has 1 X PCI-E 16 and 2 X PCI-E slots.

Yes, but you have to get cards that are appropriate for the slots you have -- you can use one x16 card and one x1 card.   Matrox has a PCIe x1 version of the Millenium G550, which you can buy here: http://shopmatrox.com/usa/products/datasheet.asp?ID=787, and there are a few nVidia Quaddro PCIe x1 cards targeted at the workstation market (i.e. they're a bit pricey).

Another alternative is to use one dual-display x16 card and this (it's not available yet, but will be in a couple weeks):

... or a couple of these:  http://www.matrox.com/graphics/offhome/dh2go/home.cfm
