Command Line script to Defragement the HDD

Our File Server is allocated 850GB (Raid 5) HDD space of SAN. I want to know how I can run a defragmentation even when the users are accessing the data. Also want to know roughly how long its going to take to defragment the 850GB HDD. I want to run this a scheduled job on every week. Any help and suggestions are appreciated

Thanks for your quick comments.

Can we tell how long is the deframentation going to take place. At the moment we have 3% of  HD being fragmented.
havent seen the ability for the defrag tool to tell how long its going to take

Would their be any performance issues or any problems if we defragment quite often.
only whilst defragging, it can slow down the system big time, its is highly advisable to do this out of hours...have a read through this

I ran chkdsk yesterday night, said that would dismount all the handles to SAN. Pressed Yes for that.THis has made SAN in accessible with for the users.
Ckdsk was taking too long so I have end the task.
Came in the morning and realized that 3 folders were corrpted. Can't delete move or rename the folders. No NTFS permissions tab at all.
you ended chkdsk partially complete? ouch! what did the folders contain?
