Did you installed the latest service pack for SQL server?  I think it is your security setting when you install the SQL server.  I usually set Authentication with "SQL Server and Windows" instead of just "Windows Only".  In your asp.net application you use the SQL Server authentication to connect to the SQL Database.

I think you forget to start the SQL Service!  You need to start the SQL service first and then go to "Property" and select the "Autostart SQL Server" checkbox under "General" tab, so next time SQL server start automatically when you start your computer.
