I would say Backup Exec, version 11d

Others would argue Arcserve.

I like BUE as it is stable, works, is very easy to use and can doo all the jobs you need. It works with practically all tape devices from single all the way up to Robotic libraries, and also works a treat with Backup to Disk which is what you are requiring.

It has remote agents that you install on the other machines to be backed up and you can select these through a directory tree, right down to individual files.

forgot to add, works a treat with Exchange Server as well, backing up either individual mailboxes or the entire Storage groups.

Version 11d also now has continuous Exchange protection.

Have a read through: http://www.symantec.com/backupexec/index.jsp

There are some nice flash animations explaining things. Maybe a bit dumbed down but it gives you a good overview.
Two big comparable backup application are Backup Exec (Symantec) and Tivoli (IBM). Backup exec is best use for backing the data to media (tape). BA is good but is not totally reliable compare to IBM Tivoli backup. Tivoli is designed to do disk backup over the network. It is faster than BA, but it's a bit pricey compare to BA. It's all depending on your need and budget. If you do need to do disk backup over the network (LAN or WAN) with healthy budget, Tivoli can be your consideration. If you backup to tape, and have a limited budget, Symantec BA is your shot.
