I have created master page and put scriptmanager in it. Then creted default.aspx and get the masterpage content. But if I want to place a button in my default page, it is saying not declared, even though it is declare on the aspx page. THis is my first experience with script manager with master page and default page relation. What I need to do?

First close all the files then open only aspx page and first remove the button from the page, save it.
Then again add a new button and name it as you wish and then Press F7 it will open the codebehind file and now check whether the button object has been created or not.
