How can I utilize Linux to protect my Windows based LAN

I have heard that you can use a linux box to 'scrub' mail and internet traffic to help protect from viruses and other 'bad' things.  If this is possible,  how does one do this and how can I get started?

Is it possible to do this:

Email in Lan -> SMTP Server -> ISP and visa versa?

Will the SMTP  server keep the emails separated so they go to the right internal mail box?
