custom member email addresses used for a domain

I have a client who wants to do this:

He has a membership site. He wants the members to each have their own email address under his domain and all mails will go out from his site to their custom address, forwarding to their real email. And then when someone hits reply, it will go back to the person using their custom email address.

How is this done? What tools/ applications are needed to do this?

My mail server is merakmail. I don't know if that can do it, but if not, what can do it?

It would have to be done via the code because there is no way I could manually add thousands of accounts a day. It is apparent you can do it through the code somehow, because craigslist does it as soon as you click the button to send, so they are accessing the mail server database via the code and setting up accounts somehow.

Also, he wants it to be anonymous so the users don't know one another's contact information, but I don't think this can ensure anonymity because the users can always type in or reply with their real email addresses in the body.

Are there any truly anonymous ways of doing this? I want to go with an online mail system and can parse for email addresses and strip them out.

not quite the answer I'd hoped for. but i want to clean this up. decided to go a different route since I couldn't get a really good answer on how to do this.
