Here are some hints for you to check...

Have a grant access to ASP.NET user (IUSR_...) to the IIS virtual directory.
Please assign an IP for the IIS and let testers to test with the IP address.
Are testers in a different domain other than the application installed?
    Any firewall blocks?

Go to Directory Security in IIS and go to Anonymous Access configuration by selecting "Edit..."

--How to assign an IP for the IIS and let testers to test with the IP address ?
Goto Web site tab in IIS properties and select IP address that you want to assign.
