Windows cannot access the specified device, path, or file

Hi,
I am working on an XP Media Center PC that has been infested with Spyware & Trojans. I have made some headway in SAfe Mode, installing and running SPYWare Doctor several times, and Hijackthis. I am still unable to do anything in Normal Mode, as evertime I attempt to run an exe, or such as regedit, or even run Control Panel, the message "Windows cannot access the specified device, path, or file" results. Any advice on how to resolve this? I suspect a registry setting or File association.

In normal Mode, the Task Manager keeps getting disabled, preventing me from shutting down suspect processes so I can work to eliminate them. I can enable it, but is is immediately dis-abled again. In Safe Mode, once I disable it, it remains dis-abled.

All help appreciated.
