Currently we dont have complex passwords in our group policy on our network.  My question is, if I enable complex passwords when will it take effect to the users?

Currently users are prompted every 42 days to change the password, will they have to make a complex password the next time they get prompted or will all users be prompted right away??

It will take effect at the next password change, so either when they are prompted or when they choose to change their password.
