how to lock a windows file from the command line (or from vba)

I am writing some server software that walks through a directory and "does things".  (Exactly what gets done is not relevant to the question).

I want to test how well the new server program works when some of the files are locked by other processes, so I want commands like these.

for instance   <c:\mydir\mylocker   "\\server01\directory to process\filename.vsd">
and later      <c:\mydir\myunlocker   "\\server01\directory to process\filename.vsd">

I wish I could simply open the file with its native application, but that will not work.  For instance, the above example uses Visio, which is not available on my computer.
Plus, there are many other file extensions I might encounter.

a command line program, or a programming technique or DLL that can be called from VBA code, or Autoit code would be best - I don't have access to VB or C++ compilers.
