This is possible. How it is build depends on how you want your configuration. Can you tell more about the desired situation? Is it just for virtual machines or should physical machines also connect through this ISA server?

If it's only for VM's you can create an extra virtual switch (internal only). This switch gets no network cards. In your virtual machine you add another virtual network card which you connect to the newly created vSwitch.

After configuring your ISA server you can move the VM's from the original vSwitch to the newly created vSwitch. Now all data travels through the ISA server.

Or you can attach on NIC to your outside connection and confirgure that on a seperate vSwitch. From there attach one of the VMs NICs to the WAN vSwitch and as well to the LAN vSwitch. From there configure your ISA server with the external IP on that NIC connected to the WAN vSwitch.
