Changing the page file location on SBS

I would like my SBS 2003 page file on E: rather than C:  I see where to change it (in system>advanced>performance, etc.), but I need to know about the "timing".  Can I simply go change it while the system is up and running with users connected?  Or, would I get everyone off and do it then, or reboot first, or go into safe mode, or what's best?

And, once I do this and control panel shows that it is running on E: and I see it on E:, is it then safe to delete it from C:?  Are there any "tricks" to getting it to delete at that time?  TIA
