In your web.config file you can create your connection string like this...
http://weblogs.asp.net/owscott/archive/2005/08/26/Using-connection-strings-from-web.config-in-ASP.NET-v2.0.aspx

For example...

<connectionStrings>
    <remove name="LocalSqlServer" />
    <add name="LocalSqlServer" connectionString="data source=.\SQLEXPRESS;Integrated Security=SSPI;AttachDBFilename=|DataDirectory|aspnetdb.mdf;User Instance=true" providerName="System.Data.SqlClient"/>
    <add name="MainConnStr" connectionString="data source=.\SQLEXPRESS;Integrated Security=SSPI;AttachDBFilename=|DataDirectory|main.mdf;User Instance=true" providerName="System.Data.SqlClient"/>
</connectionStrings>
