Are you positive of the location being in system ? instead of the system32 folder?
If so, then it was indeed a virus but now it's gone and only the loading point in the registry that is still calling for the dead/gone file to load hence the error.

In msconfig > StartUp tab you can uncheck the relevant entry to stop the error.
Or run Hijackthis and fixing the relevant 04 entry(mostly) will fix the error.

Start > Run > type in:
msconfig

click on StartUp tab, and look for the relevant entry pointing to that file.

Or, Download Hijackthis:
http://www.trendsecure.com/portal/en-US/tools/security_tools/hijackthis/download
Open Hijackthis, click "Do a system scan and save a logfile" don't fix anything yet.
Paste the log in the "Code Snippet" or "Attach File" window.
Or run a scans that cleans up remnant registry entries
