I would like to know if a software update on Mac mini from ver 10.3.9 Panther to ver 10.5.4 Leopard is possible. Should I expect any problems during the update and later on running Mac?

Many thanks for helping

You can definitely go from Panther to Leopard with little effort or expected issues.  Understand that this is a paid update since it is a change in major versions, but any store that sells Mac software should carry it.

The disc is original which has been paid for. What issues I could come across? Do I need to take any special steps beforehands?

You should always verify your necessary software works in 10.5 (haven't run into anything that won't lately) and backup any non-replaceable data before performing any major OS upgrade.
