File sharing through ISA 2006.

Can I use ISA 2006 to allow remote file sharing?
And it will be great if you can give me the best practices to do that.

You're not going to get Windows file Sharing to work over a Firewall.  There is a whole lot more to it than "ports".  For that matter you can be on the same network on the same subnet and still have trouble with Windows Filesharing,...let alone be across a firewall.

Establish a Web Server or an FTP Server and then Publishing it using the ISA.

Another option that we have also used is to install a cheap NAS. Ours is a Linksys NAS200 with a pair of mirrored Terabyte drives. It has a built in FTP & WWW service along with the ability to have its own local user accounts.  You publish it from ISA just like you would an FTP Server or a Web Server.  It is more secure because it is self contained and at the very worst a hacker is only going to screw up the NAS and not screw up the LAN

Sure you can.

The most common way is to enable the pptp VPN option within ISA - assuming you have ISA deployed with two nics rather than just as a proxy server and one nic.
