using Member Profile Builder and cannot save profile info

I have a very basic login web application that I am try to use profiles with.  I am using the Web Profile Builder since it is a Web Application rather than a Web Site.   I can retrieve and use the profile information but can't update or save any changes to the profile data.  

According to the documentation assigning to the Profile member such as
Profile.ClientNumber = TextBox1.Text

Should make the assignment,  but it is not saving any information.

What am I missing ?

did you call the Save-Method?
var user = new WebProfile().GetProfile("User");
user.City = "New city";
user.Save();
