I would highly recommend not defragmenting an exchange server while exchange services are running. This is very dangerous as it can damage your InfoStore. Are you running a nightly defragment? I would recommend scheduling a task to stop exchange during system maintenance such as a defrag, disk check or even restart.

A simple "net stop msexchangesa /y" command will work great!

Best,

Check this out:

http://www.techotopia.com/index.php/Repairing_and_Defragmenting_Windows_Server_2008_Disks

It has some nice photos. Check your schedule and disable any automatic scheduling.

When you say "goes down by defragmentation", how do you know this is the case? Is there an event for this? Is it the online defragmentation for the directory?

MS says it's a performance related issue.. can you run ExBPA and see what is the result?
are you updated the exchange with latest Roll up update?
IS your Exchange si on DC with GC?

Refer the above article and add MSExchangeSA to DependOnService at HKLM\System\CurrentControlSet\Services\MSExchangeIS. and see if the error re-appears
