I need to understand the difference between SAP IS-U and SAP R/3 and how they integrate. What is SAP IS-U used for? and what does it offer that cannot be found in standard SAP R/3?

You can think of IS-U as a set of components and settings inside the R/3 system. Along with standard SAP R/3, SAP has provided solutions that cater to the special demands of a particular Industry and are seamlessly integrated with the components of standard SAP. These are called Industry Specific solutions.

Does it mean that it adds more tables to the R/3, e.g. billing related tables? what are these? I need a bit more technical architecture of the link between IS-U and the standard R/3 thx

Yes, more tables, and other objects. I do not know exactly which tables, programs, etc, etc that is added when IS-U is installed into the standard R/3 system. But there are lot of them.

Again, think of IS-U as a set of objects that are installed info the standard R/3 system, along with the standard objects.
