Have you checked the MySQL documentation? If you are using MySQL 5.1, see the following:
http://dev.mysql.com/doc/refman/5.1/en/create-tablespace.html
http://dev.mysql.com/doc/refman/5.1/en/alter-tablespace.html

The most basic usage of a MySQL tablespace is:

CREATE TABLESPACE my_tablespace
 add datafile 'my_tablespace_file'
 engine falcon
;

CREATE TABLE `my_table`
 unique_id INT NOT NULL AUTO_INCREMENT PRIMARY KEY,
 my_column VARCHAR(10) NOT NULL
) tablespace = my_tablespace;

However if you want anything more specific you'll need to be a lot more specific in your question.
