"What operating system runs on the Buffalo Box NAS devices?"

There are several types of Buffalo NAS devices; they all run on a specific version of Linux firmware that was developed exclusively for them. Custom built variants of Debian, Fedora and Gentoo may be installed as well.

How do I determine which users are connected to the Buffalo Box NAS before I power it down?

Try to telnet into your NAS. Login with your admin account and pass. If that won't work, use "root" as login with no password (just hit ENTER).
On the Linux console, issue the command:
who
or:
who -u

In case this does not work, try to see if there is an administrative GUI that you access via a web browser (usually the IP address or hostname of the NAS plus a specific port). There ought to be a manual of some kind for details.
