My current setup is Dell r805 with Esxi and direct storage. I have the OS install to the SD card in the server.  I wanted to know if the datastore/partition is the same form esxi to esx.  Could i then just reinstall esx onto the sd? Will the esx see the datastore created in esxi?

Yes, it is the same. Both use a VMFS3 volume/partition. Your reinstalled ESX OS will 'see' the datastore. You'd have to re-add it like you did in ESXi, but it will 'see' it.
