I wear a lot of hats - Developer, Database Administrator, Help Desk, etc., so I know a lot of things but not a lot about one thing. Experts Exchange gives me answers from people who do know a lot about one thing, in a easy to use platform." -Todd S.

In my installation of WSUS the directory you are referring to does not exist.

Your Path: e:\wsus\wsuscontent\WsusContent\B1
My Path: e:\wsus\WsusContent\B1

Looks like a corrupt install, check to see if the directory is there if not create it and if that doesn't work then try a repair/reinstall of WSUS

Yeah I checked and don't have that path.  Mine is like yours.
Mandeep KhalsaCommented:
Try creating that additional "WsusContent" directory and see if it lets you download the updates.

Did you move the content ??
goto %drive%\Program Files\Update Services\Tools.
Run this command: wsusutil movecontent e:\wsus c:\wsuslog.txt -skipcopy

Check the logfile c:\wsuslog.txt to see if it was successful, then run the following from the same location:

wsusutil reset

This will check the DB against the stored location and update the tables as necessary.
