HTTP 500 Internal Server Error

I have a Windows SBS2003 server I believe to be causing  a very strange problem.

The server itself has Internet access as expected.  As of last Friday all clients within the network are unable to access external webpages, they can all ping the server, gateway and any webpage (www.google.com) also tracert to any IP or DNS name, they can all open internal webpages (companyweb) but nothing external, the "HTTP 500 Internal Server Error" message is displayed.

All clients can also open ftp.microsoft.com, just nothing http or https.

There is no firewall or ISA running on the server only a Fortigate device, I have confirmed that this device is not blocking traffic, so am sure there is something going on on the server.

There are no relevant error messages in either client or server logs to indicate a problem.
Have done a full virus scan on the server using Trend Micro SMB, which could not find any threats.

When running the Network Diagnostic tool, it comes back sayin that it could not make a http or https connection.

Any suggestions would be appreciated.

check the framework and the configuration files of the server and your website configuration

I have installed firefox on one of the clients and that works, IE still does not (Even after being reset to defaults).
I think there must be some sort of malicious program at fault here, so will investiagte that a bit further.
