IIS 6 "403 - Forbidden: Access is denied."

I'm getting this error when I try to browse to a site on IIS6.

403 - Forbidden: Access is denied.
You do not have permission to view this directory or page using the credentials that you supplied.

Path credentials is set to a specific user.
This user has the appropriate rights to the website's physical path.
In IIS, when I click on "Basic Settings" and "Test Settings", I get:
   "The specified user credentials are valid."
   "The specified user credentials are authorized to access the specified physical path."

What could I be missing?
It is an asp.net 4.0 website using "Forms" authentication.

Use Process monitor it will provide the which process is locked due to permission.

Sorry but this didn't help.
It didn't show me which process was locked due to permission.
