I wear a lot of hats...

"The solutions and answers provided on Experts Exchange have been extremely helpful to me over the last few years. I wear a lot of hats - Developer, Database Administrator, Help Desk, etc., so I know a lot of things but not a lot about one thing. Experts Exchange gives me answers from people who do know a lot about one thing, in a easy to use platform." -Todd S.

This is a very broad question.    Are you looking at troubleshooting a specific issue or looking at being a tech and looking at supporting exchange?

For both Ex2003 and ex2007, make sure to check DNS as well.

Start with the Event Viewer, check for DNS or Exchange errors.

If errorsdo occur, start troubleshooting Exchange 2003 with this tool.

For Exchange 2007 you can make use of the same tool, though not every part of the tool is Ex2007 aware.

My recommendation, which you might already be doing, is pickup a book or two and read.   A book that I'm reading now is http://www.amazon.com/Microsoft-Exchange-Server-2010-Practices/dp/0735627193/ref=sr_1_1?ie=UTF8&qid=1287495600&sr=8-1
Its good to read the books because you pick up points on the products that you might otherwise miss.    It has a chapter also on troubleshooting.   In addition to all this its typically experience and reseaching when you have an issue.
