Sys.Extended.UI and Sys is undefined, one or another

if i have <asp:ToolkitManager id="tkmg01" runat="server" CombineScript="false" ..>
it will solved the Sys.Extended.UI error,

but after i browsed to upload the file (with ajax asyncuploadfile), it has the error 'Sys is undefined'
To solved this 'Sys is undefined', i have to turn CombineScript="true", but then this will cause the first error.

workaround with
<pages enableEventValidation="false" enableViewStateMac="false" viewStateEncryptionMode ="Never" />
