what is the best way to call a C dll from Windows Server 2008?

Can I call it from a cmd or bat job?
Or do I have to use a scripting language like Perl or VBS?

You can call a router within a .dll file by using rundll32.exe.

Example: rundll32 printui.dll,PrintUIEntry /?

This calls the function or routine PrintUIEntry within the printui.dll. Anything afterward would be passed as parameters or options to the routine.
