Change ESXi 4.1 Block size

Can anyone give me a step by step the block size so I can have more thatn 256 GB for the Guest OS?  I don't see anywhere on Sphere Client that I can modified the block size.

Pretty much the only way to do this is to remove/delete your datastore, then re-add it under Config tab -> Storage -> Add Storage link. In the Add Storage wizard, configure the block size to what you need it to be:
1MB => 256GB virt disk size
2MB => 512GB virt disk size
4MB => 1TB virt disk size
8MB => 2TB virt disk size

***NOTE: when removing/deleting your datastore, you lose all data/VMs off of it, so you'll need to migrate them to a different datastore before doing this.

unfortunately, it's this simple:

copy/move all data to another LUN
delete/remove the datastore
re-create the datastore and set correct block size
copy/move back all of your data and re-register VM's if necessary.

p.s. if you have your ESX/i install on the same partition as your datastore, that's even more cumbersome
