Create your drop down list and bring up the preferences for that object. Select the "Options" tab and select "Commit selected value..." (that is, besides specifying the items). Then go to the Validation tab and select to create a custom validation script.

Use the attached script. The script assumes that the checkboxes are called CheckBox_One, CheckBox_Two and CheckBox_Three. This works for a single selection, but if your dropdown list allows multiple selected items, the whole thing gets a bit more complicated.

event.value == "One" ? this.getField("CheckBox_One").value = "Yes" : this.getField("CheckBox_One").value = "No";
event.value == "Two" ? this.getField("CheckBox_Two").value = "Yes" : this.getField("CheckBox_Two").value = "No";
event.value == "Three" ? this.getField("CheckBox_Three").value = "Yes" : this.getField("CheckBox_Three").value = "No";
