We have blocked some sites like facebook, youtube, etc and now we noticed that somebody accessing these sites with VPN softwares through the same proxy. Can any one help me to block these proxy(VPN) softwares like hotspot shield, ultrasurf, etc use in network.

The simplest way of course is to create a rule in web access layer and place it high in the rules order
The syntax would be:
source "any" - destination "combined object proxy vpn" -  service "any - action "deny"
Combined object should contain "request URL" objects for the sites you need to deny access to
To find out exactly where these apps go, use IEinspector for example - free trial - and run tests to replicate users behavior.
