First, a few questions...which version of Visual Studio are you using? Unless I'm mistaken, Visual Studio 2008 only knows about .NET Framework versions up to 3.5, not 4.0. If the website you've loaded, setup perhaps in a different environment (?), was already targeted for Framework 4.0, but is now being run in VS 2008, that could conceivably explain the problem. VS2008 would load the site, read web.config, and see that it was targeted for a version of .NET with which it isn't familiar.
More info about your environment would be helpful, too, eg OS version (and bitness), VS version, all that good stuff, will help us help you.
