On PostBack your ModalPopupExtenders will be dismissed.  You need to "reshow" them in whatever method you are using with the RequiredFieldValidators, e.g. Button_Click, if the page is not valid.

Example in C#

protected void Button_Click (object sender, EventArgs e)
{
   if (!Page.IsValid)
  {
     myModalPopupExtender.Show();
  }
}

I doubt this is an issue with the RFVs.  This is a common issue when using modal popup extenders with validators.  The validators fire, but since a postback occurs the modal popup is dismissed.  In other words, the error message is displayed, you just don't see it.
