VB .NET Manually change settings after compile for My.Settings?

After I publish and install my application (VS 2008), I locate it in \Apps\2.0\... and modify the .exe.config file that stores the settings I set up prior to publishing.  However, after I modify these it seems like the program is still using the old values.  Is it possible to change these manually?  If not, what's the best approach for this?
