While logged into vCenter, click the Summary tab; rt-click on the datastore and select 'Delete'. *MAKE SURE YOUR DATATORE IS INDEED EMPTY AS DELETING THE DS ALSO DELETES ALL DATA ON IT.

Right Click on the Datastore and select DELETE.

But do not do this if you need any VMs, on the datastore, this process is not reversible.

To delete from Array, login to GUI management, and Delete the Array.

just to underscore what the other two have already said... make sure that you remove it from within vSphere and then do a rescan on all hosts connected to the SAN.  If you pull a LUN out from underneath an ESX host prior to removing it from the host, you can get yourself into an APD (all paths down) condition where the host becomes unmanageable and VM's might crash.  The only resolution at that point is to reboot the host.  This was fixed in 4.1 U1 (not sure about 4.0, but I believe it's in the last patch release) but it doesn't affect ESX 3.x.
