Basically I cannot see where I can do this.  I am testing TMG for lab purposes at the moment as we are evaluating it.

Create a new domain sets or URL sets... and add it it into the from tab in access rule.
There are two steps to achieving what you are trying to do, which may be why you cannot find it.  The first piece is called "URL Filtering". URL Filtering does not provide and blocking mechanism itself, but merely returns a category based on the URL.  The second step relates to rules defined for the connection (Block all sites categorized as Forbidden/Porn/etc) which can block a site if it falls into a category forbidden by a rule.
When you contact a website,  url filtering dissects to the URL to see if any hits match definitions.
