I have a user on Exchange 2010 in my organization and a mailbox on Exchange 2003. Is it possible to have the Exchange 2010 users using Outlook add an additional mailbox hosted on Exchange 2003.

Also, can it work the reverse way a Exchange 2003 user add an additional mailbox on Exchange 2010?

Are you using RPC/HTTP?  If the servers are in the same exchange organization, you should be able to open additional mailboxes from either the 2003 or 2010 server both ways.  Obviously you will need to have permissions to the mailbox(es) you wish to have access to.
Grant permission. then go to outlook account settings --> more setting --> advanced and add the additional mailbox.
