I have an Acer Extensa 4220 laptop and want to re-install the OS. I do not have the original DVD and I already have the key.

Where can I download the OS? In Microsoft's website one has to buy another key in order to re-install the OS. I do not want to do that.

Thanks.

If you want to use the license provided by Acer you'll need to get the reinstallation DVD from them - it's not available as a download.  Have you tried launching the Recovery Partition (Alt+F10 when booting) to recover the system?

See Restore and Recovery in the User Manual
You can buy replacment media from Acer here
