Web server and Mail server

How does the 2 servers talk/communicate and how to optmize or best practice implement it?
Im planning to expose a form in a website .This form has 2 fields Username and email address.
When user populates this 2 fields and click on a link, Im not sure whether direct it to a mail server or web server. funcitonally,Id like to verify if the email address and username exists . Id like to check this and build a logic to respond . Id like to use a mail server for the response but the checking/verifying in the database should be done by the application which resides in a web server.
Is this the right thing to do or whats best thing in this model?

Sounds to me like the application should live on a web server and you should query / send or do whatever you need to do to the mail server across your network.  When you say:  "Id like to use a mail server for the response," do you mean responding via sending email?  Im just really not clear on what your trying to achieve here but, in best practice, its best to separate them ( code and email server ) onto 2 different servers.
