Run remove-mailbox
Then run online maintenance
Run offline defrag if the space can be claimed.

Offline defrag off the database will reclaim space but you need 110% free space to run it. This is because eseutil creates a new database leaving you with two. You can then switch exchange to the new database.

You need to really make sure your backups are working and that exchange is successfully clearing down the transaction log files. This is usually the most common reason why space is suddenly at a premium.
