This is using Symantec Backup Exec 2010 r3, on Windows 2008 server. My management wants to have a comprehensive media set backup. For example, I am using one cartridge (Media) for weekly backup for 3rd, 4th and 5th Friday. I want to keep these 3 copies for one month. After one month, this media should be wipe off and re-use from the fresh again for another month. How should I set the settings? in Media set? in Job setup? I shouldn't do the formatting manually.

This should do the trick:
1. On the media Set:
       * Append Period: 16 days
       * Overwrite Protection Period: 30 days
2. On the job:
       * Media set: Friday Weekly 03 04 05 Media Set
       * Overwrite settings: Append to media. Overwrite if no appendable media is available.
3. In Tools/Options:
        * In Media Management: Select "Overwrite recyclable media in the target media set before using scratch media".

You will need 2 media in the media set of course. One month it should use the one media, the other month the other media. (Provided the media has enough capacity to fit the backup 3 times on the tape - if not you will need more tapes).
