You can use transport rules to do all of this, have a read of this http://technet.microsoft.com/en-us/library/aa995961.aspx

Please check the below link for How To Block A User From Sending & Receiving Emails externally In Exchange 2010…

In the same way you can create Transport rules according to your specifications.

1) Block certain users/groups from sending any mail to external recipients

In hubserver -> Select Transport Rules -> New Transport Rule -> "Set Name" -> select "From People" -> specify users -> select "Delete or Send rejection"
Specify the message and error code -> Finish

2) Block certain users/groups from sending to certain internal recipients

Create DL in EMC -> Properties -> Delivery Restrictions and add the users or Groups to restict

3) Block specific files or files in specific folders from being sent by specific user to external recipients

Select Transport Rules -> New Transport Rule -> "Set Name" -> select "From People" and sent to users that or inside and outside and when an attachment contains words -> add people and specificy the attachment words -> If you have any exception "Select it" -> Finish

4) Block specific files or files in specific folders from being sent by specific users to internal recipients

Same step in 3 and only when you select "sent to users that or inside and outside organization" you have to select here internal organization.

5) Block certain internal users from ever receiving e-mails from specific  internal users

Open EMC -> Create DL -> Poperies -> add the specific users to Delivery restriction

6)  Block certain internal users from ever receiving e-mails with specific attachments

Same in step 3. select particularly when an attachment contains words

Thank you
