Add the site to your IE Trusted site and check whether there is any error.

You can  update the change to all users using Group policy in AD.
There is nothing on the exchange part to be set up for enabling and disabling cookies.

Some Group policy which might have changed your IE settings on the User workstations.
I have about 150 users login in from different part of the world, they all started to complain yesterday that they cant surf the OWA using IE? kind of weird! it was working fine the day before
I know that according to http://support.microsoft.com/kb/299331 it is a browser problem bit it is not! it was working for all the clients than suddently it stopped working!
just to add, if I type in https://exchange.mydomain.com/owa it works fine, if I type in webmail.mydomain.com which is a web forwarder with hard link to https://exchange.mydomain.com/owa I get the error message Please enable cookies for this Web site.
