Hi Team,

When i run the Test-Mapiconnectivity -server Mb1 and its generating the below error message. please suggest.

It's a new installation or migration from 2003 ?

If is a migration from 2003, check this article:

To fix the problem, load up ADSI Edit and navigate to
 
CN=Configuration,DC=yourdomain,DC=local
     CN=Services
          CN=Microsoft Exchange
               CN=YourOrganizationName
                    CN=Administrative Groups
                         CN=Your Old Exchange 2003 AdministrativeGroupName
                               CN=Servers
 
If like me, you have finished your conversion to Exchange 2010 and no longer have any Exchange 2003 servers, the Servers container should be empty.  Deleting the empty Servers container immediately fixed the Test-MAPIConnectivity problems.  DO NOT delete the old Exchange 2003 Administrative Groups as this will cause problems.  Only remove the Servers container.
 
Please use caution whenever dealing with ADSI Edit.  Best practice would be to have the AD recycle bin enabled so you could restore the container if you have any problems.
