VMware Workstation supports Virtual Serial Ports, or you could use a USB to Serial Port Converter, for use with VMware Workstation or VMware vSphere 4.x or 5.0.

We still use serial ports under VMware Workstation or VMware vSphere 4.x or 5.0 for SCADA remote monitoring applications at remote sites.

Provided your computer is NOT using an OEM License of Windows, you can create a Physical to Virtual "copy".

I would recommend and use VMware vCenter Converter Standalone 5.0

Both VMware Player and Oracle's VirtualBox allow you to assign the serial ports to the VM's, but whether they work as expected I don't know. As both of these products are free (I prefer VMware Player as it seems better in performance over VBox) I suggest you just try it out.

Whether this is possible with type one HyperVisors I don't know, but as VMware's ESXi is free too, you can also try that.

VMware Workstation and vSphere 4.x and 5.0 work fine with serial ports and USB to Serial port converters. There is no native serial port support in vSphere 4.x (ESXi) and 5.0 (ESXi).
You can also get serial to ethernet adaptors that come with virtual com port software
