You cannot upgrade from 5.0 to 5.5.  You need to upgrade first to 5.1, then upgrade to 5.5.  But since you'll be already at it, I think it's best you take the trouble and take the steps to upgrade to 5.5.  It's been around for a while now so it's stable, and you have performance and other benefits.

See http://dev.mysql.com/doc/refman/5.1/en/upgrading.html for 5.0 to 5.1 upgrade, then
http://dev.mysql.com/doc/refman/5.5/en/upgrading.html for 5.1 to 5.5 upgrade.  There are also additional Windows-specific instructions at http://dev.mysql.com/doc/refman/5.1/en/windows-upgrading.html and http://dev.mysql.com/doc/refman/5.5/en/windows-upgrading.html.

The steps will include executing commands to update your tables to the new version, but your actual data should be left intact.  Of course, as the documentation pages referred to above will tell you, it always pays to have a backup done before proceeding.
