Exchange 2010 transport rule blocking email after being deleted

We have a site where the local admin has created a transport rule that they forgot to restrict by destination email address and thus started bouncing all email for their org - not good.

We found the cause of email rejection and disabled the rule, still rejecting, deleted rule, still rejecting stopped transport service and still rejecting, rebooted server and still it rejects all inbound email even though the rule doesn't exist and it's been some 45 minutes since first deletion

Transport rule returns:
#< #5.7.1 smtp;550 5.7.1 Lianne xxxxxxx is no longer employed by the xxx. Her cases are being dealt with by Amy xxxxxx> #SMTP#

There are no rules enabled or indeed that exist on the only Exchange server that contain reference to this ex-employee
